IGF-1 LR3 (Long R3 IGF-1) is a synthetic analogue of human insulin-like growth factor-1 engineered for enhanced potency and extended biological activity. Two key modifications distinguish it from native IGF-1: a glutamic acid to arginine substitution at position 3 (Arg3), and a 13-amino-acid N-terminal extension peptide. Together, these changes dramatically reduce binding to IGF binding proteins (IGFBPs), leaving more free IGF-1 LR3 available for receptor activation.

Research Background
Enhanced IGF-1R activation – By evading IGFBP sequestration, IGF-1 LR3 has approximately 2-3x the potency of native IGF-1 in cell proliferation assays. It activates the IGF-1 receptor (IGF-1R) and downstream PI3K/Akt and MAPK/ERK signalling cascades.
Cell proliferation and survival – IGF-1 LR3 is widely used in cell culture as a growth factor supplement and in research models studying cell proliferation, anti-apoptotic signalling, and tissue growth.
Comparison with IGF-1 DES – IGF-1 DES is an N-terminally truncated IGF-1 variant (amino acids 4-70) with different pharmacokinetics. IGF-1 DES has a shorter duration but higher receptor binding affinity, making the two variants complementary research tools.
