FOX04-DRI 10 mg – Senolytic Research Peptide
FOX04-DRI is a D-retro-inverso (DRI) peptide that disrupts the interaction between FOXO4 and p53 in senescent cells. Developed by researchers at Erasmus University Medical Centre (Netherlands), it was published in Cell in 2017 as a selective senolytic, capable of triggering apoptosis specifically in senescent cells while sparing healthy cells.
Research Background
Mechanism: FOXO4-p53 disruption – Senescent cells survive by sequestering the pro-apoptotic protein p53 via FOXO4 binding. FOX04-DRI competitively disrupts this interaction, releasing p53 to trigger mitochondrial apoptosis selectively in senescent cells.
D-retro-inverso design – The DRI modification reverses the peptide backbone chirality, making FOX04-DRI resistant to protease degradation while maintaining binding specificity. This gives it a much longer functional half-life than conventional L-peptides.
Senescent cell clearance – Accumulation of senescent cells is a hallmark of ageing, contributing to chronic inflammation (“inflammaging”), tissue dysfunction, and age-related pathology. Senolytics like FOX04-DRI are studied for their ability to selectively remove these cells.
Ageing phenotype research – Published studies reported improved fitness, fur density, and renal function in aged animal models following FOX04-DRI treatment, driving significant interest in the geroscience community.
